在三乙胺的作用下,N-取代苯基-马来酰亚胺分别与α-氯代肟基乙酸和α-氯代肟基乙酸乙酯发生1,3-偶极环加成反应,合成了14种新型5-芳基-3a,6a-二氢-4,6-二氧代氮杂茂并[3',4'-d]异(噁)唑-3-甲酸衍生物(3a~3g)和5-芳基-3a,6a-二氢-4,6-二氧代氮杂茂并[3',4'-d]异(噁)唑-3-甲酸乙酯衍生物(4a~4g),其结构经1H NMR、IR和元素分析测试技术得以表征.对化合物进行了初步生物活性测试.结果表明,化合物3a、3b和3c样品浓度为10 μmoL/L时.对人白血病细胞生长的抑制率均为100%.化合物3d、3e和化合物4a、4d、4e对细胞分裂周期磷酸酯酶Cdc25A具有不同程度的抑制作用:当质量浓度为2×10-5 g/mL时,其抑制率分别为61.47%、66.23%、18.92%、22.49%和56.43%.并探讨了其构效关系.
