通过LB膜仪对表面压力-面积(π-A)曲线测量及原子力显微镜(AFM)观测,研究了胆固醇对磷脂酰胆碱/神经酰胺二元体系结构的影响.π-A曲线分析表明,随着胆固醇的增加,加速了相变的发生,促进了膜稳定状态的形成;AFM观察结果表明,磷脂酰胆碱/神经酰胺(PC/CER)二元混合系统单分子膜呈现为小的微区结构,当PC/CER二元混合系统与胆固醇混合时,随着胆固醇比例的增加,混合膜呈现从链状结构向较大的片状与网状共存结构的转化,最终纯胆固醇形成高度紧密的膜结构.

This paper investigated the effect of cholesterol on the structures of the phosphatidylcholine (PC) and ceramide(CER) of duality system by the pressure-area(π-A) isotherm measurements and by atomforce microscopy(AFM) observation. It is shown from the π-A isotherms that the increase of cholesterol can accelerate the occurrence of phase change and advance the stabilization of films. Also, it is found from the observation of AFM that the duality systems (PC/CER)molecular film takes on microregion structure. With the increasing of cholesterol content,the mixed films transforms from the chains structure to larger slice and net coexisted structure. In the end, pure cholesterol forms more aggregated structure.
