高级检索

运用硝酸钬溶液对蚕豆根尖染毒, 提取根尖细胞DNA进行琼脂糖凝胶电泳; 取根尖压片观察细胞核异常情况; 同时取根尖细胞进行单细胞凝胶电泳. 结果表明, 钬离子诱导了根尖细胞核异常; 在8 mg·L-1剂量下, 彗星细胞尾长和拖尾细胞数量在24 h内随着根尖处理时间的延长而增加, 处理24 h时, 琼脂糖凝胶电泳中出现明显的连续拖尾带型; 处理48 h后彗星拖尾缩短, 琼脂糖凝胶电泳中拖尾带型缩短, 推测可能发生了DNA修复作用或者交联作用. 64 mg·L-1剂量处理24 h, 彗尾缩短且出现核碎裂, 琼脂糖凝胶电泳中未出现拖尾带型. 实验中未观察到细胞凋亡的典型 DNA"梯状带型"和特征性彗星拖尾现象.
